5-Hydroxymethyl-2’-deoxyuridine(5-HmdU)isformedfromthymidineduringtheoxidationofDNAbyionizingrADIationorreactiveoxygenspecies(ROS)thatareformedfrominflammatoryresponse.Thiscausesgeneticmutations,alteredgeneexpressionandchromosomalinstABIlitythathavegenerallybeenimplicatedwithaging,cancersandautoimmunediseases.
Systemicoxidativestresslevels,asevidencedbyDNAdamageproductsinthebloodandurine,appeartobeassociatedwithariskofvariouscancers,includingthatofthebreast,lungandcolorectalcancers.CigarettesmokecontainsnumerouschemicalcarcinogensandothercompoundsthatgeneratereactiveoxygenspeciesthatcandamageDNAdirectlyorindirectlyviainflammatoryprocesses.5-HmdUisoneofthemajoroxidizedDNAbasesthatissignificantlyelevatedinthewhitebloodcellsofwomendiagnosedwithbreastcancer,whichmaybearesultofdiminishedDNArepair.IndividualswithBRCA1andBRCA2mutationsmaybeatincreasedriskforcancerduetodeficienciesintherepairofDNAlesionscausedbyROS.Additionally,antibodytitersrecognizing5-HmdUhavealsobeenfoundtobesignificantlyelevatedinwomenwithbreast,colorandrectalcancers.Therefore,oxidativeDNAdamageisthereforeanattractiveMarkerofdiseaseriskasittakesintoaccountnotonlytheexposuretoandproductionofoxidants,butalsothecells’DNArepairability.
