CD95,alsoknownasFASorAPO1,isa36kDacellsurfacetypeImembraneglycoproteinwithanapparentmolecularweightof44kDaonSDSPAGE.CD95isamemberoftheTNFreceptorfamily,whichincludesTNFR1,TNFR2,CD27,CD30andCD40.BindingofCD95LigandtoCD95orcrosslinkingofCD95byantiCD95monoclonalantibodiesleadstoapoptosisofCD95expressingcells.CD95belongstoasubgroupoffamilymembersthathaveadeathdomain(DD)whichcontains70aminoacidsnearthecarboxylterminalregionofthemolecule.ThebindingofadaptormoleculestothisDDisresponsIBLefortransmittingthedeathsignalforApoptosis.
StimulationofCD95resultsinaggregationofitsDD,leADIngtotherecruitmentofFADDandcaspase8thattogetherwiththereceptorformthedeathinducingsignalingcomplex(DISC).CD95anditsligandCD95L areinvolvedintheperipheraldeletionofactivatedmatureTcellsattheendoftheimmuneresponseanddefectsinthispathwaypredisposetoautoimmunedisorders.CD95isalsoinvolvedinkillingoftargetssuchasvirusinfectedcellsorcancercellsandkillingofinflammatorycellsatimmuneprivilegedsites.
