PARC(p53associatedparkinlikecytoplasmicprotein)antibodydirectlyinteractedandformedanapproximately1MDcomplexwithp53inthecytoplasmofunstressedcells.Intheabsenceofstress,inactivationofPARCinducednuclearlocalizationofendogenousp53andactivatedp53dependentapoptosis.OverexpressionofPARCpromotedcytoplasmicsequestrationofectopicp53.FurThermore,abnormalcytoplasmiclocalizationofp53wasobservedinanumberofneuroblastomacelllines;RNAinterference-mediatedreductionofendogenousPARCsignificantlysensitizedtheseneuroblastomacellsintheDNAdamageresponse.TheseresultsrevealedthatPARCisacriticalregulatorincontrollingp53subcellularlocalizationandsubsequentfunction.
