XIAPAssociatedFactor-1(XAF-1)bindstoXIAP,aninhibitorofcaspases-3,-7,and–9,andtriggersitsrelocationfromthecytosoltothenucleus.OverexpressionofXAF-1resultsintheneutralizationofXIAP’sABIlitytoinhibitcelldeath.XAF-1isnormallyexpressedinalladultandfetaltissuesbutwasfoundtobepresentinverylowlevelsinavarietyofcancercelllines.Incontrast,XIAPlevelshavebeenshowntobehighinamajorityofcelllines.LowXAF-1andhighbasalexpressionofXIAPmaythereforeplayacriticalroleinmaintainingsurvivalofcancercelllines.BothIFN-a2andIFN-bcaninduceXAF-1mRNAinallcellsexaminedbutinductionofXAF-1protein(asobservedbyimmunoblotanalysis)wasseenonlyincelllinessensitivetotheapoptoticeffectsofIFNs.
