EphrinB2,alsoknownasHtkL,ELF2,LERK5,andNLERK1(1),isamemberoftheephrinligandfamilywhichbindsmembersoftheEphreceptorfamily.Allligandsshareaconservedextracellularsequence,whichmostlikelycorrespondstothereceptorbindingdomain.Thisconservedsequenceconsistsofapproximately125aminoacidsandincludesfourinvariantcysteines.TheBclassligandsaretransmembraneproteinswhichcanbecometyrosinephosphorylateduponreceptorligation.Thecytoplasmicdomainsareapproximately80aminoacidslongandarehighlyconserved,especiallythelast33aminoacids.Severalsignalingmoleculeshavebeenshowntointeractwiththecytoplasmicregion,althoughspecificsignalingroleshaveyettobeelucidated.EphrinB2hasbeenshowntobindEphA4,EphB1,EphB2,EphB3,andEphB4(2,3).TheextracellulardomainsofmurineandhumanEphrinB2share98%aminoacididentity.OnlymembraNEBoundorFcclusteredligandsarecapableofactivatingthereceptorinvitro.Whilesolublemonomericligandsbindthereceptor,theydonotinducereceptorautophosphorylationandactivation(2).Invivo,theligandsandreceptorsdisplayreciprocalexpression(3).Ithasbeenfoundthatnearlyallthereceptorsandligandsareexpressedindevelopingandadultneuraltissue(3).TheEphrin/Ephfamiliesalsoappeartoplayaroleinangiogenesis(3).
