Nervegrowthfactor(NGF)wasinitiallyisolatedinthemousesuBMAndibularglandoverthreedecadesagoasa7Scomplexcomposedofthreenon-covalentlylinkedsubunits,alpha,beta,andgamma.ItisnowknownthatboththealphaandgammasubunitsofNGFaremembersofthekallikreinfamilyofserineproteases,whilethebetasubunit,calledbeta-NGFor2.5SNGF,exhibitsalltheBIOLOGicalactivitiesascribedtoNGF.Thehumanproteinsharesapproximately90%identityattheaminoacidlevelwithbothmouseandratbeta-NGF.
NGFisawellcharacterizedneurotropicproteinthatplaysacriticalroleinthedevelopmentofsympatheticandsomesensoryneuronsintheperipheralnervoussystem.Inaddition,NGFcanalsoactinthecentralnervoussystemasatrophicfactorforbasalforebraincholinergicneurons.NGFhasalsobeenshowntohavebiologicaleffectsonnonneuronaltissues.NGFismitogenicforafactordependenthumanerythroleukemiccellline,TF1NGFhasbeenfoundtoincreasethenumberofmastcellsinneonatalratsandtoinducehistaminereleasefromperitonealmastcells.NGFwillenhancehistaminereleaseandstronglymodulatetheformationoflipidmediatorsbybasophilsinresponsetovariousstimuli.NGFwillalsoinducethegrowthanddifferentiationofhumanBlymphocytesaswellassuppressapoptosisofmurineperitonealneutrophils.Theseresults,takentogether,suggestthatNGFisapleiotropiccytokinewhich,inadditiontoitsneurotropicactivities,mayhaveanimportantroleintheregulationoftheimmunesystem.
