CD130,alsoknownasgp130,isatypeItransmembranesignalingmoleculeof130-140kDathatisfoundoncellsurfacesusuallyassociatedwiththespecificreceptorchainsforthefollowingcytokines:IL-6,IL-11,leukemiainhibitoryfactor(LIF),OncostatinM,ciliaryneurotrophicfactor(CNTF)andcardiotrophin-1(CT-1)(1).TheoverlapofBIOLOGicalactivityofthesecytokinesisproofofthecommonrolethatgp130/CD130playsinthesignalingcascadeforthesemolecules.Theextracellularstructureofgp130/CD130iscomposedofsixfibronectintypeIIImodules.Asolubleformofgp130/CD130existsinserumandcanactasanantagoNISTformoleculesthatinteractwithgp130/CD130.CD130hasnointrinsickinaseactivity,however,itdoesappeartopossesssomeimportantbiologicalpropertiesthatrequireitspresenceduringtheearlystagesontogeny.Micethathaveamutatedgp130genesurviveonlyafewweekslargelyduetomyocardialmalformations(2).TheCD130moleculeformshomodimersinthepresenceofIL-6andIL-11receptors(3,4).Incontrast,CD130formsheterodimerswhenpresentwithLIFR,OSMR,CNTFRandCT-1R(5).AlthoughCD130doesnotappeartodirectlybindthesecytokines(OSMbeingtheonlyexception),itspresenceinconjunctionwiththecytokinespecificreceptorchainimpartshighaffinitybindingcharacteristicstothereceptors. MostcellsexpressCD130,however,theligandspecificreceptorsassociatedwithCD130displayamuchmorerestrictedcellularexpressionpatternultimatelydeterminingthecytokine-specificresponse.
