HIF-1isanuclearproteininvolvedinmammalianoxygenhomeostasis.Thisoccursasaposttranslationalmodificationbyprolylhydroxylation. It isaheterodimercomposedofHIF-1alphaandHIF-1betasubunits.Bothsubunitsareconstantlytranslated.However,undernormoxicconditions,humanHIF-1alphaishydroxylatedatPro402orPro564byasetofHIFprolylhydroxylases,ispolyubiquinated,andeventuallydegradedinproteosomes.Underhypoxicconditions,thelackofhydroxylationpreventsHIFdegradationandincreasestranscriptionalactivity.Therefore,theconcentrationofHIF-1alphaincreasesinthecell.Incontrast,HIF-1betaremainsstableundereithercondition.HIFhydroxylasesprovideinsightintohypoxiccellresponses,whichmaybeusedtohelpisolatetherapeutictargets.
