Immunogen: Purified, E. coli-derived, recombinant mouse interleukin 6 (rmIL-6)

Interleukin 6 (IL-6), also known as interferon-beta 2, 26-kDa protein, B cell stimulatory factor-2 (BSF-2), hybridoma/plasmacytoma growth factor, hepatocyte stimulating factor, cytotoxic T cell differentiation factor, and macrophage-granulocyte inducing factor 2A (MGI-2A), is a multi-functional protein that plays important roles in host defense, acute phase reactions, immune responses, and hematopoiesis. IL-6 is expressed by a variety of normal and transformed cells including T cells, B cells, monocytes/macrophages, fibroblasts, hepatocytes, keratinocytes, astrocytes, vascular endothelial cells, and various tumor cells.
It is upregulated by IL1, TNF, PDGF, IFN beta, TNF alpha, NGF, IL17 and downregulated by glucocorticoids IL4, TGF beta. IL6 appears to be directly involved in the responses that occur after infection and cellular injury, and it may prove to be as important as IL1 and TNF alpha in regulating the acute phase response. IL6 has also been implicated in regulating adipose mass.
It also plays a role in the differentiation of Nerve Cells.
Images: Figure 1-Mouse IL-6 stimulates the proliferation of the IL-6-dependent murine plasmacytoma cells, T1165.85.2.1 in a dose-dependent manner (Nordan, R.P. and M. Potter, 1986, Science 233:566 - 569). The ED 50for this effect is typically 0.02 - 0.06 ng/mL.
Figure 2-To measure the ability of the antibody to neutralize the bioactivity of rmIL-6 on T1165.85.2.1 cells, rmIL-6 was incubated with various concentrations of the antibody for 1 hour at 37° C in a 96 well plate. Following this preincubation period, T1165.85.2.1 cells were added. The assay mixture in a total volume of 100 μL, containing antibody at the concentrations indicated, rmIL-6 at 0.25 ng/mL and cells at 2 x 105 cells/mL, was incubated at 37° C for 72 hours in a humidified CO2 incubator. Resazurin was added during the final 16 - 20 hours of incubation to measure cell growth. The relative fluorescence was then read in a fluorescent plate reader set at Ex. 544/Em. 590. The ND of the antibody is approximately 0.01 - 0.03 μg/mL.
