LEF1andTCFaremembersofthehighmobilitygroup(HMG)DNAbindingproteinfamilyoftranscriptionfactorswhichconsistsofthefollowing:Lymphoidenhancerfactor1(LEF1),TCellFactor1(TCF1),TCF3andTCF4(1).LEF1andTCF1wereoriginallyidentifiedasimportantfactorsregulatingearlylymphoiddevelopment(2)andactdownstreaminWntsignaling.LEF1/TCFbindtoWntresponseelementstoprovideadockingsiteforβ-catenin,whichtranslocatestothenucleustopromotethetranscriptionoftargetgenesuponactivationofWntsignaling(3).LEF1/TCFproteinsaredynamicallyexpressedduringdevelopmentandaberrantactivationoftheWntsignalingpathwayisinvolvedinmanytypesofcancersincludingcoloncancer(4,5).
TCF4,alsoknownasTCF7L2,isexpressedwidelyduringdevelopment.GenetargetingstudiesindicatethatTCF4isrequiredtomaintainthecryptstemcellsofthesmallintestine(6,7).TCF4hasseveralsplicingisoformsthatareexpresseddifferentiallyintissuesandduringcancerprogression(8,9).StudiesalsoindicatethatavariantoftheTCF4geneconfersanincreasedriskoftype2diabetes(10).
