Tumorproteinp53(TP53),anuclearprotein,playsanessentialroleintheregulationofcellcycle,specificallyinthetransitionfromG0toG1.Itisfoundinverylowlevelsinnormalcells,however,inavarietyoftransformedcelllines,itisexpressedinhighamounts,andbelievedtocontributetotransformationandmalignancy.p53isaDNA-bindingproteincontainingDNA-binding,oligomerizationandtranscriptionactivationdomains.Itispostulatedtobindasatetramertoap53-bindingsiteandactivateexpressionofdownstreamgenesthatinhibitgrowthand/orinvasion,andthusfunctionasatumorsuppressor.Mutantsofp53thatfrequentlyoccurinanumberofdifferenthumancancersfailtobindtheconsensusDNAbindingsite,andhencecausethelossoftumorsuppressoractivity.AlterationsoftheTP53geneoccurnotonlyassomaticmutationsinhumanmalignancies,butalsoasgermlinemutationsinsomecancer-pronefamilieswithLi-Fraumenisyndrome.
