The14.3.3familyofproteinswereoriginallydiscoveredasprominentproteinspotson2dimensionalgels.Theyareafamilyof28-33kDaproteinswhichareoftenmajorproteinsofthecytoplasm(1).Theyfunctionasbindingpartnersforphosphoserineandphosphthreoninesitesinotherproteins,thoughtheyhaveotherbindingpartnerswhicharenotdependentonphosphorylation.ThesebindinginteractionsareimportantintheregulationofmoleculessuchasthesignalingkinasesintheMAPkinasepathwayc-Rafandb-Raf,theproapoptoticmoleculesBadandBaxandthecellcycleregulatorCdc25.
