Neurofilamentscanbedefinedastheintermediateor10nmfilamentsfoundinspecificallyinneuronalcells.Intheelectronmicroscopeneurofilamentsappearsas10nmdiameterfibresofindeterminatelengthwhichgenerallyhavefinewispyprotrusionsfromtheirsides.Theyarefoundparticularlyabundantlyinaxonsoflargeprojectionneurons.AntibodiestothevariousneurofilamentsubunitsareveryusefulcelltypeMarkerssincetheproteinsareamongthemostabundantofthenervoussystem,areexpressedonlyinneurons,andarebiochemicallyverystable.

MammalianNF-Hcontainstandemrepeatsbasedonthesequencelysineserineproline(KSP),whichmaybepresentupto60times,withsomevariABIlitybetweenspecies.TheseKSPsequencesarethesitesofphosphorylation,andinaxonsmostifnotalloftheserineresiduesarephosphorylated.Incontrast,neurofilamentsindendritesandperikaryaarenotphosphorylatedontheseKSPsequences.Axonalneurofilamentsarethereforequitedifferentimmunologicallyfromthoseinperikaryaanddendrites,andmanyantibodiesstainonlyaxonaloronlydendritic/perikaryalneurofilaments.Sincebacteriadonotphosphorylatemammalianproteins,thisproteinresemblestheNF-Hformcharacteristicofperikaryalanddendriticneurofilaments.Accordinglythisantibodystainsdendriticandperikaryalneurofilamentsparticularlywell.
