Calbindin,alsoknownascalbindin1orcalbindin-D28kisamemberofthelargesuperfamilyofcytoplasmicCa2+bindingproteins.Calbindin-1belongstothesubclassofthesecontainingthe"EFhand"Ca2+bindingmotiforiginallycharacterizedinparvalbumin.Calbindinisexpressedinmammalianbrain,intestine,kidneyandpancreas.Inthebrainitislocalizedincertainclassesofneurons,andantibodiestoitareusefulforidentifyingspecificneuronalcelltypes.ItisparticularlyconcentratedinthedendritesandperikaryaofcerebellarPurkinjecells,butisalsofoundinmanyGABAergicinterneuronsinthecortex.

Thefunctionofcalbindin-1appearstobeprimarilybufferingtheCa2+ levelincells.TheaffinityofcalbindinforCa2+ islowatthetypicalrestingcytoplasmicCa2+ levelofaround100nM,andtheproteinonlybindsCa2+ significantlywhenlevelsincreasegreatly.Accordingly,itiswidelythoughtthattheprimaryfunctionofthisproteinistoactasaCa2+ buffer.BufferingCa2+ isimportant,asuncontrolledincreasesinthelevelofthiscationcanleadtobothapoptosisduetoCa2+stimulatedreleaseofproteinsfrommitochondriaandnecrosisduetotheactivationofCa2+dependentproteases.
