CalretininisamemberofthelargesuperfamilyofcytoplasmicCa2+bindingproteins,thusbelongstothesubclassofthesecontainingthe“EFhand”Ca2+bindingmotiforiginallycharacterizedinparvalbumin.Calretininisexpressedinmammaliancentralnervesystem,testis,fallopiantubeandpancreas.Inthebrainitislocalizedincertainclassesofneurons,andantibodiestoitareusefulforidentifyingspecificneuronalcelltypes.Itisparticularlyconcentratedinsomecerebellargranularcellsandtheirparallelfibers,butisalsofoundinmanyGABAergicinterneuronsinthecortex.TheseGABAergicinterneurons,inmostcases,expressonlyoneofthreeCa2+bindingproteins,namelycalretinin,calbindinorparvalbumin.Asaresult,theseimportantinhibitoryinterneuronscanbeidentifiedandsubclassifiedbasedontheircontentofthesethreeproteins.Eachtypeofneuronasdefinedinthisfashionhasparticularelectrophysiologicalandfunctionalproperties.Forexample,calbindinpositiveinterneuronsarenotfast-spikingasareparvalbuminexpressinginterneurons.Humancalretininisalsoknownas29kDacalbindinandcalbindin-2,foritssequenceisrelatedtocalbindin.
