Oct4,alsoknownasPOUdomaintranscriptionfactor,Octomerbindingtranscriptionfactor3andOct3,isexpressedinhumanandmousetotipotentembryonicstemandgermcells.Itmayfunctioninpreventingthesecellsfromdifferentiating.Eachtypeofestablishedmammalianpluripotentstemcellline(embryonalcarcinoma(EC)cells,embryonicstem(ES)cellsandembryonicgerm(EG)cells)expressesOct4,whichdisappearsrapidlywhenthecellsdifferentiate.MoststudiesonOct4havebeenperformedonmurineembryos.Inhumanoocytesofvariousmaturityandnormallydevelopingpreimplantation/cleavageembryos,avariableOct4expressionpatternhasbeenrevealed,concomitantwithapurecytoplasmiclocalizationoftheprotein.ThisvariABIlityinexpressionfadesawayduringcompaction,indicatingembryonicOct4expression;theproteinappearsinthenucleus,implyingBIOLOGicalactivity.
FunctionalOct4hasbeenfoundinhumanamnioticfluid.AsOct4transcriptshavealsobeenfoundinvariousdifferentiatedcells,thepresenceofOct-4transcriptsorproteinsmaynotbesufficientforidentifyingundifferentiatedcelllinesinhumans.ItmayalsobeimportanttoexaminethelocalizationofOct4proteinswithinacell.Oct4hasbeenshowntointeractwithNanogandwithproteinsfrommultiplerepressioncomplexes,includingtheNuRD,Sin3AandPmlcomplexes,possIBLytocontrolESfate.ReactivationofOct4expressionispostulatedtooccurindifferentiatedcellsthathaveundergonecarcinogenesis,ortumorformation
