Visualarrestinwasfirstdiscoveredasaresultoftheexperimentalmodelofhumanuveitis,anautoimmunediseaseoftheeye.Inthismodel,calledexperimentalallergicuveitis,animalswereinjectedwithextractsmadefromtheretinaofthesamespeciesmixedwithFreund’scompleteadjuvant.Theanimalsmountedastrongimmuneresponsetotheextract,andtheantibodyresponsewasusedtoidentifyseveralimmunogenicretinalproteins.OneofthesewascalledS-antigen,forsolubleantigen.Theproteinwasfoundtobeabundantinretina,about48kDainmolecularweight,andlocalizedintheoutersegmentsofthephotoreceptors.
Severalyearslater,HermannKühnandcolleaguesdiscoveredthatthisproteinbindstophosphorylatedrhodopsinandpreventsthisproteinfromactivatingtransducin.TransducinisatypicalheterotrimericGprotein,composedofαandβγsubunits.RhodopsinphosphorylationismediatedbyRhodopsinkinase(a.k.a.GRK1),theprototypicmemberofafamilyofGPCRkinases.SincetheS-antigenproteinarrestedtheactivityofrhodopsinitwasrenamedarrestin,andbecametheprototypicmemberofthearrestinproteinfamily.
