TwospecificcellsurfacereceptorsthatbindIGF-IandIGF-IIhavebeenidentified.ThetypeIIGFreceptorthatparticipatesinIGFsignalingisstructurallyrelatedtotheinsulinreceptor.Itisadisulfide-linkedheterotetramerictransmembraneglycoproteinwithanintracellulartyrosinekinasedomain.TypeIIGFreceptorbindsIGF-IwithhigheraffinitythanIGF-II.ThetypeIIIGFreceptorwhichbindsIGF-IIwithmuchhigheraffinitythanIGF-Iisalsothecation-independentmannose6-phosphatereceptor.Atthepresenttime,itisnotknownifthetypeIIIGFreceptorparticipatesintheIGFsignalingpathway.AnadditionalunknownreceptorwhichmediatesIGF-IIsignalinghasalsobeenproposed.CirculatingIGFsexistincomplexesboundtoIGFbindingproteins.Currently,atleastsixhighaffinitybindingproteinshavebeenidentified.
