LeukemiaInhibitoryFactor(LIF)isalymphoidfactorwhichpromoteslong-termmaintenanceofembryonicstemcellsbysuppressingspontaneousdifferentiation.LIFhasanumberofotheractivitiesincludingcholinergicneurondifferentiation,controlofstemcellpluripotency,boneandfatmetabolism,mitogenesisofcertainfactordependentcelllinesandpromotionofmegakaryocyteproductioninvivo.RatLIF(rtLIF)isa19.9kDaproteincontaining183aminoacidresiduesthatexhibits91%aminoacidsequenceidentitywithmurineLIF1.AnalysisofthesecondarystructureofratandmurineLIFindicatesadifferenceinthetwodimensionalstructureofbothproteins1.StudieshaveshownthatrtLIFismoreeffectiveinmaintainingtheundifferentiatedphenotypeofratEScellsthansimilarconcentrationsofmurineLIF1.
