Fibronectinisahigh-molecularweight(~440kDa)extracellularmatrixglycoproteinthatbindstomembrane-spanningreceptorproteinscalledintegrins.[1]Inadditiontointegrins,fibronectinalsobindsextracellularmatrixcomponentssuchascollagen,fibrinandheparansulfateproteoglycans(e.g.syndecans).Fibronectinexistsasadimer,consistingoftwonearlyidenticalmonomerslinkedbyapairofdisulfidebonds.Thefibronectinproteinisproducedfromasinglegene,butalternativesplicingofitspre-mRNAleadstothecreationofseveralisoforms.
MeasuredbyitsABIlitytosupportcellattachmentandspreADIngwhenusedasasubstratumforcellculture.Inthisapplication,therecommendedconcentrationforthiseffectistypically1-5μg/cm2.Fibronectincanalsobeaddedinthemediatosupportcellspreadingataconcentrationof0.5-50μg/mL.Optimalconcentrationdependsoncelltypeaswellastheapplicationorresearchobjectives.
