ThehumanFGF-9CDNAencodesa208aminoacidresidueproteinthatcontainsasingle,potentialN-linkedglycosylationsite.Thenativeproteinisglycosylatedandisefficientlysecretedaftersynthesis,althoughFGF-9lacksatypicalsecretionsignal.RatandmouseFGF-9showaveryhighhomologytohumanFGF-9.ThetranscriptsforFGF-9havebeenfoundinbrainandinkidneytissue.FibroblastGrowthFactor-9isamemberofthefibroblastgrowthfactor(FGF)family.FGFfamilymemberspossessbroadmitogenicandcellsurvivalactivities,andareinvolvedinavarietyofBIOLOGicalprocesses,includingembryonicdevelopment,cellgrowth,morphogenesis,tissuerepair,tumorgrowthandinvasion.FGF9wasisolatedasasecretedfactorthatexhibitsagrowth-stimulatingeffectonculturedglialcells.Innervoussystem,thisproteinisproducedmainlybyneuronsandmaybeimportantforglialcelldevelopment.ExpressionofthemousehomologofthisgenewasfoundtobedependentonSonichedgehog(Shh)signaling.Micelackingthehomologgenedisplayedamale-to-femalesexreversalphenotype,whichsuggestedaroleintesticularembryogenesisFibroblastGrowthFactor9mayhavearoleinglialcellgrowthanddifferentiationduringdevelopment,gliosisduringrepairandregenerationofbraintissueafterdamage,differentiationandsurvivalofneuronalcells,andgrowthstimulationofglialtumors.
